钙咪唑啉对人胃腺癌MGC-803细胞增殖分化的影响

【摘要】 目的:探讨钙调蛋白在癌细胞增殖分化中的作用。方法:应用钙调蛋白拮抗剂—钙咪唑啉(CalmidazoliunCDZ)50~150nmol/L作用于人胃腺癌MGC803细胞,用白细胞计数法观察到CDZ对人胃癌细胞的增殖有明显的抑制作用,利用3HTdR掺入、PDE活性测定、CAMP含量测定、流式细胞光度术、cAMPPDEase电镜细胞化学技术进行系统研究。结果:钙调蛋白拮抗剂CDZ抑制MGC803细胞的DNA合成,一定浓度的CDZ阻断MGC803细胞于G1期,经CDZ处理的癌细胞内磷酸二脂酶(PhosphodiesterasePDE)活性下降,并使细胞内cAMP水平瞬时升高,癌细胞质膜上的3’.5’环磷酸腺苷磷酸二酯酶(cAMPPDEase)活性明显下降。结论:上述结果表明钙调蛋白在人胃癌细胞的增殖分化过程中起重要的调控作用,有理论和实际应用的价值。

【Abstract】 Objective:To study the effects of calmidazolium(CDZ),a calmodulin antagonist,on the proliferation and differentiation of human gastric cancer cell line MGC 803.Methods:MGC 803 cells were treated with CDZ (50~150nmol/L)and cell proliferation was evaluated by survival cell count;DNA synthesis were analyzed by 3H TdR incorporation and flow cytometry assay;cAMP level and phosphodiesterase(PDE)activity were determined by radioimmunoassay and cytochemical staining.Results:CDZ strongly inhibited proliferation of MGC 803 cells and DNA synthesis,it brought about a block at the G1 phase of MGC 803 cell cycle.CDZ decreased the activity of PDE and increased cAMP level in MGC 803 cells.Conclusion:The results suggest that the growth inhibition of MGC 803 cells by CDZ was related with the regulatory mechanism of cAMP and calmodulin.
