随机扩增多态性DNA技术对淋球菌耐药相关性分析

【摘要】 目的 :用随机扩增多态性DNA (RandomAmplifiedpolymorphismDNA ,RAPD)技术分析淋球菌DNA ,评价不同菌株间的克隆相关性及耐药性传播。方法 :对 3 0株临床分离的淋球菌进行了 β 内酰胺酶检测 ,用K -B法进行了12种抗生素药敏试验 ,以随机引物OPA -0 3对 3 0株菌DNA进行RAPD分析。结果 :经OPA -0 3扩增淋球菌DNA ,琼脂糖凝胶电泳EB染色 ,最多可获得 8个不同大小的DNA片段 ( 2 0 0~ 2 0 0 0bp) ,不同菌株扩增出 1~ 8个片段 ,3 0株淋球菌可获得 5种不同的RAPD指纹图 ;扩增产物经聚丙烯酰胺凝胶电泳银染色最多可获得 15个不同大小的DNA片段( 2 0 0~ 2 0 0 0bp) ,不同菌株扩增出 8~ 15个片段 ,3 0株淋球菌同样可获得 5种不同的RAPD指纹。结论 :具有相同耐药谱的不同菌株有不同的DNA多态性 ,DNA多态性相似或相同的菌株间其耐药谱相近或相同 ,虽然两种电泳染色结果各有优缺点 ,但对淋球菌菌株间克隆相关性及耐药性传播分析结果是一致的 ,表明RAPD有助于了解菌株间的克隆相关性及耐药性传播

【Abstract】 Objective:To evaluate the clonal relation and spread of antibiotic resistance of Neisseria gonorrhoeae by random amplified polymorphism DNA (RAPD).Methods:30 Neisseria gonorrhoeae isolates,tested for β lactamase and susceptibility to 12 antibiotic by disc agar diffussion method,were characterised by RAPD analysis with arbitrary primer OPA 03.Results:After RAPD with arbitrary primer OPA 03,8 different DNA fragments from 200 to 2000bp were obtained by agarose gel electrophoresis with EB stain among the 30 isolates and 1~8 fragments with different sizes could be obtained from the strains.The 30 isolates were classified into 5 RAPD patterns;and 15 different DNA fragments from 200 to 2000bp were obtained by polyacrylamide gel electrophoresis with silver stain among the 30 isolates and 8~15 fragments with different sizes could be obtained,the 30 isolates were classified into 5 RAPD patterns as well.Conclusion:Strains shared common antibiotic profile may had different DNA polymorphism.Strains shared similar or common DNA polymorphism had similar or common antibiotic profile.Although the two electrophoresis methods had different advantages and disadvantages,the conclusions were agreeable.So it can be concluded that RAPD is helpful for the investigation of clonal relation among strains and spread of antibiotic resistance.
