一种新型低压离子色谱柱填料的研究

【摘要】 对一种新型低压离子色谱柱填料—球形离子交换羧甲基纤维素进行了研究。它具有亲水性及大孔开放性结构 ,可在低压下操作。采用梯度洗脱 ,UV检测 ,可以实现蛋白质的分离以及二肽和三肽的分离。

【Abstract】 A novel packing material in low pressure ion chromatography was discussed in the paper.It was spherical cellulose ion exchanger which was prepare with synthetic spherical cellulose and some organic compounds by suspension method.Its partical size was 30~100 μm.Among them 30~50 μm partical was used for analytical parking material and 50~10 μm panrical was used for preparetive parking material.the spherical cellulose ion exchanger was especially used for separating biosystem such as proteins and peptides as sationary phase duo to its hydrophilicity,special operture and open construction in 101325 Pa low pressure. Proteins were separated on it and were determinated with UV detector.Proteins were separated with 0.2mol/L CH 3COOH-CH 3COONa.buffer (A)and 1mol/L NaCl solution(B) as mobile phase with gradient elution Flow rate A:0.7?ml/min.B:0.3?ml/min.Dipeptide and tripeptide were separated with 0.2?mol/L CH 3COOH-CH 3COONa buffer.Flow rate was 0.8?ml/min.The best separation was obtained when the column was 5?mm×100?mm.
