酶联免疫吸附法测定乳粉中IgG含量

【摘要】 母乳化乳粉中添加保健功能因子人IgG。用饱和硫酸铵法从健康人血清中制备粗IgG ,经SephadexG -1 0 0柱制备纯IgG ;用纯人IgG作为抗原 ,免疫兔子制备兔抗人IgG抗体。建立乳制品中人IgG含量酶联免疫吸附测定 ,最佳包板物浓度为 5.2 5μg/ml,最佳血清效价为 1∶1 60 0 ,抗血清亲和力为 2 .4 0× 1 0 7,回收率为 88%~ 94 % ,测定IgG浓度范围为 5.0~1 60ng/ml,最小检测限为 3 .2 5ng/ml。

【Abstract】 A human Ig*#G was incorporated into the humanized milk powder. A rough human Ig*#G from the soundy humanserum was extracted by saturated ammonia sulfate,then the rough human Ig*#G was pured through sephadex G-100 column. The antibody of antihuman Ig*#G was made with pured human Ig*#G injected in rabbit. A enzymelinked immunosorbet assay of human Ig*#G content in humanized milk powder was developed, with a optimal concentration of coated micropore plate of 5.25*!μg/ml,a optimal titration of antihuman Ig*#G serum of 1∶1*!600,a affinity of antihuman Ig*#G of 2.40×107,the recovery of 88%~94%,a detecting concentration of human Ig*#G in 5.0~160*!μg/ml and a minimal limite of detecteing concentration of 3.25*!μg/ml.
