植物谷胱甘肽磷脂氢过氧化物酶基因的筛查
Screening of phospholipid hydroperoxide glutathione peroxidase genes in plants
【摘要】 为了解谷胱甘肽磷脂氢过氧化物酶 (PHGPx)基因在植物中的分布情况 ,以 10种具有重要经济价值的植物为对象 ,采用 PCR法和 Southern杂交法进行 PHGPx基因的筛查 ,结果显示 ,从来自萝卜、藿香、芦荟、番茄、苦瓜、小麦、油菜和大蒜的 DNA 样品中获得了 PCR扩增片段和Southern杂交阳性带 ,表明这 8种植物中存在 PHGPx基因 ,并初步确定其在基因组中的拷贝数为 1~ 2 ,为了解植物中 PHGPx的分布及克隆这些基因提供了基本信息
【Abstract】 The distribution of plant phospholipid hydroperoxide glutathione peroxidase (PHGPx) genes was investigated by screening conducted using the PCR and southern blotting methods in ten important plant species. The results demonstrated the presence of PCR fragments and southern hybridization positive bands produced from the DNA samples of radish, wrinkled giant hyssop, aloe, tomato, bitter gourd, wheat, rape and garlic suggesting that the PHGPx gene may exist in each of these eight plant species with 1 or 2 copies in their genomes. These results provided basic information for cloning these genes.
