分步电泳法检测古糖酯
Detection of Propyleneglucol Guluronate Sodium Sulfate(PGSS) with Discontinuous Electrophoresis
【摘要】 应用分步电泳法检测水溶液和大鼠尿液中的古糖酯 (Propyleneglucol Guluronate SodiumSulfate,PGSS) .该醋酸纤维膜电泳选用两种电泳液 :(1) 0 .1mol· L-1的醋酸锌溶液 ,p H6 .0 ;(2 )1.0 mol· L-1的醋酸钡溶液 ,p H5.0。电泳膜于 0 .5%阿利辛兰水溶液中染色 ,1%醋酸溶液褪色。该法能够检测不同介质中的古糖酯 ,水溶液中的检测下限为电泳膜上加样 10 0 ng,尿液中古糖酯浓度高于 50 0 ng/ m L,经提取可以用此法测得。实验证实该法灵敏、稳定性高、重现性好。
【Abstract】 To detect propyleneglucol guluronate sodium sulfate(GPSS) in solution and pooled rat urine with cellulose membrane discontinuous electrophoresis, two electrophoretic buffers were employed:0.1 mol·L -1 zinc acetate and 1.0 mol·L -1 barium acetate. The resulting electrophoretic stripe was stained by 0.5% alcian blue and destained by 1% acetic acid. As low as 100ng GPSS loaded on the membrane and 500ng/mL of GPSS in pooled rat urine were detectable with this method. The detection has been proved to be sensitive, reliable and reproducible.
