卵泡直径和卵丘细胞对牛卵母细胞体外受精后发育潜力的影响
Effects of Follicle Size and Cumulus Cells on Developmental Competence of Bovine Oocytes Following Maturation, Fertilization and Culture in vitro
【摘要】 本实验以屠宰场牛卵巢为材料,研究了卵泡大小和卵丘细胞对卵母细胞体外受精后发育潜力的影响,结果表明卵泡大小和卵丘细胞直接影响卵母细胞受精后的发育。直径为2~8mm卵泡中的卵母细胞经体外成熟和体外受精后,卵裂率(60.9% VS 37.0%, P<0.05)和受精卵的囊胚发育率(56.0% VS 38.0%,P<0.05)明显高于直径小于2mm卵泡中的卵母细胞。而直径在8mm以上的卵母细胞虽卵裂率显著低于2~8mm卵泡中的卵母细胞(45.7 %,P<0.05),但囊胚发育率无显著差异(49.3%,P>0.05)。来源于2~8mm卵泡、周围没有卵丘细胞的裸露卵母细胞经成熟培养和体外受精后,卵裂率为41.9%,但52.1%停留在2~7-细胞期,最高发育阶段为16-细胞期。当裸露的卵母细胞与来自同类卵泡的卵丘细胞卵母细胞复合体(Cumulus Oocyte Complexes, COCs)共培养20~22h后,虽然卵裂率没有明显提高,但有17.5%的受精卵能发育到桑椹和囊胚阶段,这表明COCs在成熟过程中释放促进典母细胞成熟的物质。卵母细胞与卵丘细胞间的完整联系对提高卵母细胞成熟后的受精率和囊胚发育率非常重要,有部分卵丘细胞的
【Abstract】 This experiment was carried out to study effects of follicle size and cumulus cells on developmental competence of bovine oocytes derived from abattoir ovaries following maturation, fertilization and culture in vitro (IVMFC). The results showed that both follicle size and cumulus cells directly affected development of oocytes after IVMFC. Both cleavage and blastocyst rates of oocytes from follicles which diameters were 2-8 mm were significantly higher than from follicles which diameters were less than 2 mm (60.9 % VS 37.0 %; 56.0 % VS 38.0 %, P<0.05). Although the cleavage rate of oocytes from follicles which diameters were more than 8 mm was significantly lower than from follicles which diameters were 2-8 mm (45.7 % VS 60.9 %, P<0.05), the blastocyst rates didn’t exist difference between two groups (56.0 % VS 49.3, P>0.05). Oocytes without cumulus cells (naked oocytes) from the follicles which diameters were 2-8 mm could cleavage following IVMFC. The cleavage rate was 41 .9 %, but most embryos (52. 1%) stopped at 2- 7 cell stage and the highest stage was at 16-cell. However, some embryos (17.5 %) could develop into morulae and blastocysts after naked oocytes were co-cultured with cumulus oocyte complexes (COCs) for 20-22 h. It implied that COCs might secreted some factors to promote the maturation of naked oocytes during its maturation. It was very important for obtaining higher cleavage and blastocyst rate atter IVMFC that oocytes were completely surrounded by cumulus cells. The cleavage rate of oocytes surrounded by part cumulus cells was significantly higher than naked oocytes (52.9 % VS 44.4 %, P<0.05) following IVMFC. Both cleavage and blastocyst rates of oocytes surrounded by more than 3-laper cumulus cells were significantly higher than oocytes with part cumulus cells (62.9 % VS 52.9 %; 54.5 % VS 18.1 %, P<0.05).
