猪肥胖基因(ob)位点的RFLP多态性分析
RFLPS Polymorphic Analysis on Obese (ob) Gene Locus in Swine
【摘要】 肥胖基因(ob)产物—Leptin是反映体内脂肪含量和调节体重的重要信号因子。本研究从构建的猪脂肪cDNA文库中分离克隆猪ob基因,并以克隆的猪ob基因片段为探针,Bg/Ⅱ酶切基因组DNA对不同品种猪在ob基因位点的多态性进行了分析。结果表明脂肪型猪与瘦肉型猪在RFLP带型上存在明显差异,在所有瘦肉型猪中都存在一条4.3kb的带,脂肪型猪存在一条3.5kb的带,在所分析的6个品种,共60个样品中检测到4种基因型的存在。
【Abstract】 Obese gene (ob) is a new gene cloned recently. The product of ob gene-leptin is an important signal retlecting body fat mass and controlling body weight. Here we report the cloning of pig ob cDNA gene and the polymorphic analysis on ob locus in swine. Using the cloned pig ob cDNA 1.5 kb fragment as probe and restriction enzyme Bgl Ⅱ to digest genomic DNA, we analyzed the Polymotphism on ob locus in swine. The results indicated that the RFLP band types are clearly different between fat-type and lean-type pig breeds. All the lean-type pigs provided a 4.3 kb band, and all the fat-type pigs give a 3.5 kb band. We detected 4 kinds of genotypes in all the samples analyzed so far, which included total 60 samples from 6 pig breeds.
