人精子膜甘露糖结合蛋白的研究——蛋白的定位
A study of mannose binding protein on human spermatozoan membrane——the protein location
【摘要】 作者运用生物素标记的山羊抗人巨噬细胞甘露糖受体 (GoatAnti humanMacrophageMannoseRecep tor ,AMMR)IgG对人获能精子膜上的甘露糖结合蛋白进行定位 ,经激光共聚焦显微镜分析结果显示 :AMMRIgG组阳性率显著高于运用正常山羊 (NormalGoat,NG)IgG的对照组 :AMMRIgG组的阳性率大约在 70 % (6 9.41% )左右 ,而NGIgG组小于 10 % (9.2 4% )。AMMRIgG组中 ,荧光结合部位主要在精子的顶体后区、赤道区及颈部 ,而NGIgG对照组中除少量精子显微弱的荧光外 ,绝大多数呈阴性结果。精子膜上甘露糖结合蛋白的定位研究可能对受精过程中精卵识别机制的了解具有重要意义。
【Abstract】 The fluoresecence microscopy with laser scanning confocal microscope (LSM) revealed that goat anti macrophage mannose receptor (AMMR) IgG indeed bound to the surface of living capacitated spermatozoa.The binding were mainly on the postacrosomal region,equatorial segment and neck of the human capacitated spermatozoa,where as the binding for normal goat IgG showed non specific.The mannose binding protein on human spermatozoan membrane perhaps is one kind of critical molecules which are involved in initial procedure of sperm egg recognition during fertilization.
