抑白血病因子对荷白血病鼠肝、脾细胞超微结构的影响
EFFECTS OF ANTI LEUKEMIA FACTOR ON ULTRASTRUCTURE OF LIVER, SPLEEN CELL IN BEARING LEUKEMIA MICE
【摘要】 目的 :探讨抑白血病因子对荷白血病鼠肝、脾组织及癌细胞超微结构的影响。方法 :纯系 6 15小鼠为实验动物 ,分三组 :a .阴性对照组 ,b .阳性对照组 ,c.治疗组 ,以抑白血病因子治疗。切除肝脾组织用戊二醛固定 ,以常规法制备电镜样品 ,透射电镜下观察三组小鼠肝、脾组织及癌细胞超微结构变化。结果 :阴性对照组肝、脾组织呈现正常结构 ,阳性对照组肝、脾组织结构异常 ,且有大量白血病细胞浸润 ,细胞超微结构以线粒体 ,内质网和核糖体变化突出。治疗组肝、脾组织结构趋于向正常转化 ,可见到脾窦内含有大量的癌细胞凋亡。结论 :抑白血病因子具有抑制白血病细胞增殖和浸润的作用。其作用机制可能与抑白血病因子促进白血病细胞分化 ,诱导白血病细胞凋亡有关
【Abstract】 Objective:To study the effects of anti leukemia factor (ALF) on ultrastructure of bearing leukemia mice. Methods:Inbred 615 mice for experimental animals were divided into three groups:a.negative control group,b.positive control group and,c.treatment group which was injected ALF. The liver and spleen of mice were excised and fixed with glutaraldehyde, then ultramicrotomy was progressed by routine method. Ultrastructure of liver, spleen and cancer cell were observed under transmission electron microscope. Results:The cell structure of negative control group in liver and spleen appeared normal. The cell structure of positive control group was abnormal which was invaded typically by leukemia cell and there was obviously changed the mitochondrion、endoplasmic reticulum and ribosome. The cell structure of treatment group was tended towards normal transformation. The large number of apoptosis of cancer cell was observed in splenic sinusoid. Conclusion:Anti leukemia factor have the effect of inhibitory proliferation and invasion to leukemia cell. The mechanism of anti leukemia of ALF correlated with promotion differentiation and inducing apoptosis to leukemia cell.
