浅蓝菌素对人结肠癌裸鼠移植瘤bcl-2及bax基因表达水平的影响
Influence of Cerulenin on the Expression of Bcl-2 and Bax Genes in Experimental Colonic Carcinoma
【摘要】 目的 :通过观察人结肠癌LoVo细胞裸鼠移植瘤bcl 2及bax基因表达水平 ,探讨脂肪酸合酶抑制剂浅蓝菌素选择性的抑瘤途径和机制。方法 :建立人结肠癌裸鼠移植瘤模型 ,浅蓝菌素每公斤体重 160mg腹腔注射 ,1次·d-1,连续10d ,第 17天处死并解剖动物 ,瘤组织作免疫组织化学检测bcl 2及bax基因表达水平。结果 :浅蓝菌素处理组结肠癌LoVo细胞移植瘤bcl 2基因蛋白表达率为 68.5 % ;对照组bcl 2蛋白阳性表达率为 87.8% ,阳性细胞弥漫分布。两组间有显著性差异 (P <0 .0 1)。bax基因蛋白在浅蓝菌素组移植瘤组织阳性表达率为 75 .4% ,主要呈强阳性表达 ;对照组阳性表达率为 9.5 %。两组间比较有统计学差异 (P <0 .0 1)。结论 :浅蓝菌素可使结肠癌LoVo细胞移植瘤的bcl 2基因表达下调 ,bax基因表达增强 ,提示bcl 2及bax基因在浅蓝菌素诱导的人结肠癌LoVo细胞凋亡中可能起着重要的调控作用。
【Abstract】 Objective To investigate the antitumor mechanisms of cerulenin through determination of bcl?2 and bax oncoprotein in transplantated tumor of colonic carcinomas in nude mice.Methods The expression rate of bcl?2 and bax oncoprotein in the transplantation tumor were detected by using SP immunohischemistry staining method.Results The expression rate of bcl?2 oncoprotein were significantly lower in cerulenin?treated group than that of the tumor control group.The expression of bax was contrary to bcl?2 oncoprotein.Conclusion The mechanism of cerulenin induced cell death might be correlated with apoptosis, and bcl?2 and bax gene might play an important role in regulating cerulenin?induced apoptosis.
