牛IgG Fc受体Ⅲ的分子克隆和序列分析
Molecular cloning and sequence analysis of bovine IgG Fc receptor typeⅢ
【摘要】 目的研究牛IgGFc受体的结构与功能的关系。方法在构建了牛肺巨噬细胞cDNA文库的基础上,用PCR方法获得了编码牛IgGFc受体(boFcγR)的全长编码基因。结果通过分子克隆和序列分析,发现其编码基因为507bp,共编码168个氨基酸,其中含有信号肽序列、穿膜区和胞内结构域以及3个N-连接糖基化位点。与COLLINS等克隆的boFcγR相比,在它的胞外区中缺失了82个氨基酸,仅有一个胞外结构域。结论我们所克隆的cDNA片段属于编码牛的FcγRA(CD16-)一种特殊的基因结构形式
【Abstract】 ObjectiveIn order to investigate the structural and functional relation of bovine IgG Fc receptor.Methods The full length of gene encoding Fc receptor type Ⅲ for bovine IgG(boFcγRⅢ,CD16)) was obtained with PCR from the cDNA library of bovine alveolar macrophage.ResultsIt is shown through molecular cloning and sequen cing that the coding gene has 507 bp and codes for 168 amino acids.There are gene sequence coding signal peptide,transmembrane and intracellular regions.It also has 3 potential N linked glycosylation sites.Compared with the boFcγRⅢ cDNA cloned by COLLINS et al,the sequence deletes 82 amino acids and has only one extracellular domain.ConclusionIt is suggested that this cDNA fragment may be a special form of the gene structure of bovine FcγRⅢA(CD16 Ⅱ). [
