白介素-8、肿瘤坏死因子在猪肾缺血再灌注损伤中的作用研究
STUDY ON THE EFFECTS OF CELLULAR FACTOR ON ISOLATED SWINE RENAL INJURY BY ISCHEMIA AND REPERFUSION
【摘要】 目的 :探讨白介素 - 8、肿瘤坏死因子在肾缺血再灌注损伤中的作用。方法 :采用离体猪肾缺血再灌注损伤模型 ,实验分缺血再灌注 (IR)组和对照 (control)组。检测在肾缺血 5 0± 5分钟后 ,自体稀释抗凝血再灌注 2小时 30分钟时肾组织中白介素 - 8(IL - 8)、肿瘤坏死因子 (TNF -α)的含量 ;光镜和电镜下肾组织形态结构改变。结果 :IR组同对照组比较 ,IL - 8和TNF -α均增高 ,差异具有显著性 (P <0 0 5 ) ;IR组肾组织细胞发生变性、坏死改变 ,对照组肾组织超微结构正常。结论 :IL - 8、TNF -α的增高参与了缺血再灌注性肾损伤。
【Abstract】 Objective:To investigate the effect of cellular factor on the renal ischemia-reperfusion injury. Method: The animals in the experiment were divided into ischemia reperfusion(IR) group and control group in 22 swine. After 50±5 minutes ischemia the swine kidney was cyclically perfused for 2 hours and 30 minutes with diluent blood in IR group, lipid Interleukin-8 (IL-8) and Tumor necrosis factor-α (TNF-α) in kidney tissues were detected in IR group and control group, and necrotic tubules were counted under light microscope. The ultrastructure of renal tissue was observed under electromicroscope in the two groups. Results: There were significant differences in the above indexes in comparison of the IR group with the control group(P<0.05); the degenerative and necrotic changes were remarkable under electromicroscope in renal tissue in IR group, but no change was found in the control. Conclusion: The rise of IL-8 and TNF-α was involved in the renal ischemia-reperfusion injury.
