脑梗死小鼠凋亡神经元及脑血管内皮细胞超微结构的观察
Long term observation on ultrastructure of apoptosis of neuron and endothelial cells of cerebral vessels in mice with photochemically induced cerebral infarction
【摘要】 观察脑梗死后不同时间点脑组织的病理变化 ,尤其是凋亡神经元及血管内皮细胞超微结构的改变。方法 采用经皮光化学脑梗死模型 ,氯代三苯基四氮唑 (TTC)染色 ,光镜及透射电镜观察。结果 缺血 2 4~ 48h ,脑组织及血管内皮细胞损伤最严重 ,缺血 6h至 1个月均可见凋亡神经元 ,其中以 48h最多见。早期应用巴曲酶对缺血脑组织及血管内皮细胞有保护作用。结论 脑缺血后神经元在相当长的一段时间内均可发生凋亡。保护血管内皮细胞、抑制神经元凋亡对改善缺血脑组织的预后具有重要意义。
【Abstract】 Objective To investigate the cerebral pathological changes at different time points after brain infarction,especially the changes of the ultrastructure of apoptosis of neuron and vascular endothelium.Methods The transdermically photochemically induced cerebral infarction model was used.TTC(2,3,5-triphenyl-tetrazolium chloride) staining,light and transmission electron microscopy were chosen to investigate the changes.Results The damage of brain tissue and vascular endothelium was most prominent from 24 to 48 hours after brain infarction.The apoptosis of neuron could be found from 6 hours to 1 month after brain infarction,and was found most frequently at 48 hours.The administration of batroxobin protected the brain tissue and endothelium at the early stage of cerebral ischemia.Conclusion The apoptosis of neuron could occur during a rather long period and the time window of inhibiting ischemic apoptosis is relative long.The pathomorphism of ischemic brain could be seriously influenced by endothelium.
