发根农杆菌A4转化黄瓜获得发状根再生植株

【摘要】 研究以黄瓜 ( Cucumis stativus)栽培品种津研四号为试材 ,比较了培养基中添加激素对自然生根及发状根诱导的影响 .无菌苗子叶及下胚轴经发根农杆菌 ( Agrobacterium rhizogenes) A4 侵染后 2周诱导出发状根 ,发状根在无激素的 MS筛选培养基上能够自主生长 ,在附加 BA2 .0 m g/L,NAA0 .2 mg/L 的筛选分化培养基上分化出不定芽 ,GUS染色表明转化植株呈阳性反应

【Abstract】 The experimental material in this study was cucumber culitivar Jinyan 4.The effect of addition of plant hormone in medium on natural rooting and hair y root induction was compared.Cotyledons and hypocotyl segments were infected b y A.rhizogenes A 4 and hairy roots were induced two weeks later after infec tion. Hairy roots could grow spontaneously on MS selection medium without plant hormon e.Adventitious shoots were observed on selection medium with BA 2.0 mg/L,NAA 0.2 mg/L.Histochemical analysis for Gus gene indicated that transformed plants were positive.
