新型聚乙烯醇系亲和吸附剂的制备及其在糖蛋白纯化中的应用II.吸附剂制备
STUDY ON PREPARATION OF POLYVINYL ALCOHOL AFFINITY ADSORBENT AND ITS APPLICATION FOR PURIFYING GLYCOPROTEIN ( II ) -Absorbent Preparation
【摘要】 利用已合成的聚醋酸乙烯酯-甲基丙烯酸缩水甘油酯-甲基丙烯酸烯丙酯大孔共聚物,醇解后与6-氨基正己酸反应,进一步在EDAC的作用下实现与间氨基苯硼酸的偶联,得到亲水性的亲和吸附剂;配基密度为0.865mmol/g干树脂,偶联效率为89%。以云芝糖肽作为目标物质,吸附容量为53.7mg/g干树脂,为醇解前的7.7倍。
【Abstract】 In this study, the alcoholysis reaction of macroporous copolymer Vinyl acetate-Glycidyl methacrylate-Allyl methacrylate with methanol solution leading to a more hydrophilic polyvinyl alcohol matrix was investigated, and then the affinity adsorbent was prepared using 6-Aminocaproic acid(ACA) as a spacer and 3-Aminophenylboronic acid(APBA)as a ligand in the existence of EDAC catalyst. The effects of ACA amount used, temperature and time on the addition reaction between the epoxy group of matrix and the amino group of ACA were examined. In this way, the conjugation rate of preparation course was up to 89 percent. In purifying Polysaccharide-peptide of Coriode vorsicolor (PSP) the adsorption capacity of this adsorbent is 7.7 times higher than that of the blank matrix.
