锌试剂与赖氨酸相互作用的吸光光度研究
ABSORPTIOPHOTOMETRIC STUDY ON THE REACTION BETWEEN ZINCON AND L LYSINE
【摘要】 研究了邻 {2 - [α- (2 -羟基 - 5-磺基苯偶氮 ) -亚苄基 ]-肼基 }苯甲酸 (锌试剂 ,ZCN)与 L-赖氨酸 (L- Lys)复合物的吸光光度测定基本条件 ,在 p H 1 .9的缓冲溶液中 ,ZCN与 Lys结合生成紫红色复合物 ,最大吸收波长在 60 5nm处 ,标准工作曲线在 5.47× 1 0 -6~ 1 .64× 1 0 -4 mol· L-1范围内呈线性关系。在最大吸收峰处比锌试剂本身红移 1 45nm,复合物的表观摩尔吸光系数 ε为 2 .5×1 0 4 。同时 ,研究了离子强度对反应体系的影响 ,ZCN作为光谱探针与 Lys的结合数
【Abstract】 In this paper, a absorptiophotometric study on the rection of Zincon with L lysine is reported. Zincon combines with L Lys to give a purple product, in a buffer solution of pH 1.9. The bathochromic shift of the product is about 145nm. The apparent molar absorptivity is 2.5×10 4L·mol -1 ·cm -1 at 605nm. The linear range of standard cure is 5.47×10 -6 ~1.64×10 -4 mol·L -1 . It is found that ionic concentration of the solution has significant effect on the staining reaction. The composition of zincon L lysine complexe was determined by molar ratio method.
