核酸—槲皮素—锌(Ⅱ)三元配合物荧光法测定核酸
Spectrofluorometric Determination of DNA with the Ternary of Quercetin, Zn(Ⅱ) and DNA
【摘要】 在室温条件下 ,热变性小牛胸腺 DNA与槲皮素 - Zn( )形成三元配合物 ,使槲皮素 - Zn( )二元配合物荧光大幅增强 ,激发波长 2 13nm,发射波长 4 72 nm ,基于此对核酸进行测定 ,方法简单、快速、灵敏 .p H=8.3时 ,线性范围 (0 .5~ 4 .0 ) g/ m L,检测限 0 .0 5g/ m L,线性相关系数γ为0 .9992 ,且共存物质干扰小 .
【Abstract】 A fluorometric method ha s been developed for a rapid determination of DNA with quernary Zn(Ⅱ) as a fluo rescence probe, based on the fluorescence enhancement of quernary Zn(Ⅱ) in the presence of thermally denatured DNA. Maximum fluorescence is produced in the pH range of 8.2~8.5, with maximum excitation and emission wavelengths at 213 and 4 72 nm, respectively. Under optimal conditions , the calibration graph is linear over the range of 0.5~4.0 μg/mL and the corresponding detection limit is 0.05 μg/mL for calf thymus DNA. And the influence of foreign substances is little.
