初治卵巢癌组织MDR1表达水平的定量测定及其意义
Study on the expression of a human multidrug resistance gene in ovarian carcinoma and its practical value
【摘要】 目的:探讨初治卵巢癌组织多药耐药基因(MDR1)表达水平及其对相关药物抗肿瘤作用的影响.方法:用逆转录酶链聚合反应(RT—PCR)法对37例初治卵巢癌组织MDR1表达水平进行了定量测定.结果:37例卵巢癌组织均有MDR1表达,其中浆液性癌的表达水平为0.75±0.22,粘液性癌为1.03±0.28,内膜样癌为0.57±0.11,3种病理类型之间MDR1的表达水平有显著性差异(P<0.05);34例卵巢癌的小鼠肾包膜下移植肿瘤药敏测定(SRCA)肿瘤药敏试验成功,此34例的MDR1表达水平与ADM及VCR在SRCA中的抗肿瘤作用呈负相关,与VP16的抗肿瘤作用无相关关系.结论:初治卵巢癌存在固有的MDR1表达,MDR1的表达降低相关药物的抗肿瘤作用.
【Abstract】 Objective:To investigate the expresson level of MDR1 in Ovarian Carcinoma and its effection on the antitumor ac-tivity of MDR1 reltated drugs. Methods: The MDR1 gene expression in fresh ovarian carcinom spemens from 37 patients were analyzed by reverse transcription polymerase chain reaction, and the antitumor activity of three MDR1 related drug (ADM. VCR. VP16) was mesaurde by subrenal capsule assay. Results; The MDR1 expression was demonstrated in all 37 case of ovarian carcinomas; The MDR1 /? actin in serious Carcimoma was 0.75 ?0.22, in mucious carcinoma 1.03 ?0.28, in endometrioid carcinoma 0.57 ?0.11, there were significant differences among the three histological subtypes(P<0.05); Negative correlations were found between the MDR1 expression level (specially in high) and the antitumor activity of ADM and VCR in subrenal capsule assay. Conclusion:There are intrinsic MDR1 expression in all untreated ovarian carcinomas, and the MDR1 expression may lead the chemoresistance of MDR1 related drug. Therefore, MDR1 related drugs should combined with MDR1 reverse agents and MDR1 unrelated drugs in the overian carcinoma treatment at the very begining.
