【摘要】 目的　动态观察兔眼小梁切除术中与术后应用 5 -氟尿嘧啶 (5 - Fu)后滤道瘢痕组织变化 ,探讨两种方法抑制瘢痕组织的差异。方法　对 2 4只新西兰大白兔双眼行小梁切除术。右眼术后前 10日 ,每日结膜下注射5 - Fu 5 mg/ (2 5 m g/ ml× 0 .2 m l) ,左眼术中小梁组织切除前 ,用 5 - Fu(浓度 2 5 mg/ ml)浸泡的棉片直接置于巩膜瓣下 5分钟。术后观察眼压、滤泡和并发症 ;应用光镜、电镜及计算机图像分析技术观察术后 14、2 1、2 8天滤过道瘢痕组织变化及新生胶原的相对含量 ,以及观察角膜内皮细胞和睫状体无色素上皮细胞的病理变化。结果　 (1)术中与术后应用 5 - Fu均能降低兔眼压 ,无显著性差异 (P>0 .0 5 )。两组滤泡得分无显著性差异 (P>0 .0 5 )。 (2 )术中与术后应用 5 - Fu均能抑制成纤维细胞的增殖、分泌胶原的功能 ,无显著性差异 (P>0 .0 5 )。(3)术中组与术后组的并发症无显著性差异 ,睫状体无色素上皮及角膜内皮细胞无明显毒性反应。结论　 5 - Fu注射液术中应用与术后应用抑制滤过道瘢痕组织的增生无明显差异。更多还原

【Abstract】 Objective To compare the effectiveness of intraoperative 5-Fu and postoperative 5-Fu used adjunctively with trabeculectomy in inhibition of proliferation scar tissue in rabbit.Methods Trabeculectomy were performed on both eyes of 24 rabbits.5-Fu 5mg were subconjunctivally injected postoperatively in the right eyes,as postoperative group,once daily for ten days,while a piece of cotton soaked in 5-Fu (25mg/ml) were placed under the scleral flap of the left eyes,as intraoperative group,for 5 minutes before the trabecular tissue were excised.Intraocular pressure (IOP),bleb survival and complications were analyzed after surgery.The scar tissue in filtering site,corneal endothelial cell and ciliary body epithelial cell were pathologically observed,and quantitation of the new proliferation collagen was evaluated by using microscope and electric biomicroscope and computer image evaluation technique on the 14th、21st、28th day postoperatively.Results 1.The IOP were decreased from baseline during the postoperative days 1 through 28 in the two groups.The bleb in the two groups weren’t failure until the postoperative 18 the day.There weren’t statistically significant differences between the two groups regarding the IOP and the bleb ( P >0 05) 2.5-Fu could inhibit fibroblast proliferation and collagen deposition in two groups without statistically significant differences ( P >0 05) 3 The differences of postoperative complication weren’s statistically significant in the two groups ( P >0 05).There were no harm to the corneal endothelial cell and the ciliary body epithelial cell.Conclusion Using 5-Fu intraoperatively was as effective as that postoperatively on inhibition proliferation scar tissue,therefor,it was suggested that intraoperative 5-Fu be a viable alternative to postoperative 5-Fu without subconjunctival injections conveniently.更多还原
