【摘要】 目的 :探讨细胞周期素 D1 及其激酶抑制蛋白 p15表达在急性白血病 (AL)中的临床意义。方法 :用间接免疫荧光标记流式细胞术研究 46例初治 AL 细胞治疗前后 D1 、p15、细胞周期及 DNA倍体与疗效间关系。结果 :2 8例急性髓系白血病 (AML)中 2 2例 p15阳性 ,6例阴性。 18例急性淋巴细胞白血病 (AL L)中 p15均阳性。AML与 AL L D1 均阳性。高 p15者 G1 期细胞百分率 (G1 % )增多 ,S期细胞百分率 (S% )减少。低 p15者 G1 %减少 ,S%增多。 DNA非整倍体者均高表达 p15和 D1 ,完全缓解率低 (30 % )。高 p15低 D1 者 2 0例中 18例 (80 % )完全缓解 (CR) ;低 p15高 D1 者 10例 ,8例 (80 % ) CR;高 p15高 D1 者 14例中 10例 (71.4% ) CR,低 p15低 D1 者 2例均 CR。结论 :p15和 D1 在 AL 中表达有异质性。高 p15者显示 G1 期阻滞 ,低 p15显示 G1 / S期转化 ,p15和 D1表达有助于指导治疗更多还原

【Abstract】 Objective:To study the clinical significance of cyclin D 1 and its kinase inhibitor p15 expression in acute leukemia(AL).Method:Using indirect immunofluorescence labelling,the expression of D 1 and p15 ,cell cycle,DNA ploidy and their relation to therapeutic effect were studied by flow cytometry in 46 cases of AL de novo.Result:In 28 AML cases, 22 were positive for p15,and 6 negative.All of 18 ALL cases showed positive for p15,All AML and ALL cases expressed D 1. The percentage of cells in G 1 phase increased and that of cells in S phase decreased in cases with high p15.the percentage of cells in S phase increased and that in G1 phase decreased in cases with low p15. Both high expression in p15 and D 1 occurred in DNA an euploidy cases with low complete remission rate (CR,30%).In 20 cases with high p15 and low D 1,16(80%) entered CR; in 10 with low p15 and high D 1,8(80%) CR; in 14 with both high p15 and D 1,10( 71.4 %) CR, and in 2 with both low p15 and D 1, 2 CR.Conclusion:The expression of p15 and D 1 showed heterogeneity. High p15 cases exhibited G 1 arrest ,while low p15 expression rendered the cell cycle transition from G 1 to S phase . Their expression is helpful to design therapeutic regimen.更多还原
