G-CSF动员同胞供体外周血干细胞的研究
A study of allogeneic peripheral blood stem cell mobilization with G-CSF
【摘要】 目的 :研究粒细胞集落刺激因子 (G- CSF)动员异基因外周血干细胞 (PBSC)的动力学 ,分析移植物细胞成分 ,探讨供者年龄、性别对动员效果的影响。方法 :40个 HL A相合的同胞供者一日两次皮下注射 G- CSF5μg/ kg,于第 5、6天采集 PBSC,所得 PBSC以流式细胞仪分析免疫表型。结果 :1G- CSF动员过程中供者耐受良好 ,外周血白细胞和单个核细胞 (MNC)峰值在第 5天 ;2 PBSC含有 MNC 5 .8(3.5~ 7.5 )× 10 8/ kg,CD34 + 细胞7.7(2 .0~ 16 .8)× 10 6 / kg,CD3+ 细胞 3.90 (2 .2 3~ 6 .2 8)× 10 8/ kg,CD4+ 细胞 1.98(1.2 0~ 3.14)× 10 8/ kg,CD8+ 细胞 1.4(0 .5~ 2 .5 )× 10 8/ kg。 3单次采集后血小板从 (142 .9± 46 .9)× 10 9/ L 降至 (99.8± 2 7.4)× 10 9/ L,两者有显著性差异 (P <0 .0 5 ) ;4不同年龄及性别组之间 ,MNC和 CD34 + 细胞的产率均无显著性差异 (P >0 .0 5 )。结论 :G- CSF 5μg/ kg一日两次皮下注射能有效动员 PBSC,供者的年龄、性别对动员效果无显著影响。
【Abstract】 Objective:To study the kinetics of G CSF mediated allogeneic peripheral blood stem cell (PBSC) mobilization, analyze the cellular compositions of PBSC grafts, determine the influence of donor age or gender on mobilization efficiency.Method:Forty HLA identical sibling donors were given subcutaneous G CSF, 5μg/kg twice daily . PBSC were collected on day 5,6 of G CSF administration and flow cytometric immunophenotyping were performed.Result:(1) G CSF administration was well tolerated, resulting in significant increase of peripheral WBC counts and MNCs with peak on day 5. (2) The median numbers of cells in PBSC grafts were: MNC 5.8 ( 3.5 ~ 7.5 ) ×10 8/kg,CD34 + cells 7.7 ( 2.0 ~ 16.8 ) ×10 6/kg, CD3 + cells 3.90 ( 2.23 ~ 6.28 ) ×10 8/kg, CD4 + cells 1.98 ( 1.20 ~ 3.14 ) ×10 8/kg, CD8 + cells 1.4 ( 0.5 ~ 2.5 )×10 8/kg.(3) The platelet count fell from ( 142.9 ± 46.9 )×10 9/L to ( 99.8 ± 27.4 )× 10 9/L after single collection (P< 0.05 ). (4) In terms of MNC or CD34 + cell yield per liter of blood processed, there was no significant difference between groups with different ages or genders (P> 0.05 ).Conclusion:A 5μg/kg twice daily G CSF administration schedule was efficient in allogeneic PBSC mobilization. Donor age or gender has no prominent effect on mobilization efficiency.
