VM26和手术对胶质瘤分泌β-微量蛋白的影响

【摘要】 目的:观察鬼臼霉素(VM26)和手术对胶质瘤细胞分泌β-微量蛋白(β-tp)水平的影响。方法:用ELISA检测不同剂量的VM26对体外培养的三种不同类型胶质瘤细胞培养液中β-tp水平以及测定手术前后胶质瘤病人脑脊液中β-tp的浓度。结果:①0.3~30μg·ml-1VM26对三种胶质瘤细胞增殖均有抑制作用,培养液中β-tp的水平亦降低。②少突胶质瘤和星形胶质瘤病人术后脑脊液中β-tp的水平低于术前,而髓母细胞瘤病人脑脊液中β-tp的浓度无变化。结论:VM26对胶质瘤细胞的增殖有抑制作用;手术切除少突胶质瘤和星形胶质瘤可使病人脑脊液中β-tp浓度降低。

【Abstract】 Aim: To observe the effects of VM26 and operation on beta-trace protein (β-tp ) secreted by glioma cells. Method: The levels of beta-trace protein was detected in the cultures solution of three types of glioma cells treated with doses of 0. 3~30 μg/ml VM26 were measured by the method of ELISA. By using the same method, the concentrations of beta-trace protein was detected in the CSF taken from the patients with gliomata before and after operation. Results: (1)The proliferation of glioma cells was obviously inhibited by treatment of VM26, and the concentration of β-tp was also declined in the culture solution. (2)The concentrations of β-tp in the CSF taken from patients with oligodendroglioma and astrocytic glioma after op- eration was lower than that of before operation. No changes were found in the level of β-tp in the CSF taken from patients with medulloblastoma. Conclusion: The proliferation of glioma could be inhibited by treatment of VM26. and the oncotomy could decrease the levels of β-tp in the CSF of the patients with gliomata.
