益肾化浊注射液对高糖培养肾小球系膜细胞表达FN及PAI-1 mRNA的影响

【摘要】 观察高糖对体外培养肾小球系膜细胞表达纤维连接蛋自 (FN)、纤溶酶原激活物抑制物(PAI 1)mRNA以及细胞增殖、分泌FN的影响 ,同时观察益肾化浊注射液对高糖引起上述各指标变化的影响。采用体外培养肾小球系膜细胞方法 ,用高糖及益肾化浊注射液作为刺激物 ,四甲基偶氮唑盐微量酶比色法 (MTT)检测细胞增殖 ,酶联免疫吸附测定法 (ELISA)测定细胞上清FN含量 ,Northem杂交检测FN、PAI 1mRNA表达水平。结果表明 ,高糖可促进分泌FN ,上调PAI 1mRNA的表达 ,明显抑制系膜细胞的增殖 ;而益肾化浊注射液可缓解高糖所引起的上述指标的改变 ,并可下调FNmRNA的表达。

【Abstract】 We examined the impact of YSHZ injection on the genes expressions of fibronectin(FN) and plasminogen activator inhibitor 1(PAI 1) mRNA in human glomerular mesangial cells incubated in high glucose.The expressions of fibronectin and PAI 1 mRNA were determined by northern blot analysis.The level of fibronectin in the mesangial cell supernatant was assayed through the sandwich ELISA method.The results showed that YSHZ injection could significantly antagonize the increased mRNA expressions of fibronectin and PAI 1 of the mesangial cells stimulated by high glucose.The fibronectin level in the superatant was also decreased by YSHZ.YSHZ injection could have a synergistic effect on correcting the disorder of extracellular matrix metabolism induced by high glucose.
