表皮细胞无血清培养基血清代用品的实验研究
An Experimental Study of Culturing Keratinocytes in Serum-free Media with Serum Substitute
【摘要】 目的 :进一步调查无血清培养基血清代用品中因子对体外无血清培养的人表皮细胞生长增殖的影响。方法 :利用L16(4 5)正交试验对无血清培养基血清代用品 (SerumSubstitute ,SS)中五因子的四种不同浓度进行调查 ,用MTT法测定它们分别的效应。结果 :SS中除Chol作用可疑外 ,BSA、Cata、Ins、TF均能明显促进表皮细胞的生长增殖 ,它们较好的组合是BSA 15mg/ml、Cata 10 μg/ml、Ins 8μg/ml、TF 5 μg/ml。结论 :SS在体外无血清培养基中能替代血清较好的促进表皮细胞的生长
【Abstract】 Objective: To Probe the effects of five factors contained in SS on the growth of human keratinocytes(HK) in SFM. Methods: The orthogonal trial of L 16 (4 5) was used to investigate four different densities of five factors of SS and the MTT method was used to determime their effects. Results: SS could promote growth of HK.Of them,BSA,insulin,TF and catalase were shown to have significant effect on promoting growth and proliferation of keratinocytes,,while the effect of cholesterol was doubtful. A better formula of the five factors was found to be BSA 15 mg/ml,catalase 10 μg/ml,insulin 8 μg/ml,TF 5 μg/ml. Conclusion: SS can substitute for serum to well promote the growth of HK in SFM.
