疟原虫抗药性的遗传学研究进展

【摘要】 疟原虫产生抗药性是疟疾防治中遇到的主要难题之一。抗叶酸类抗疟药的抗药性机制已基本搞清 ,与其作用靶酶二氢叶酸还原酶或二氢蝶酸合成酶基因点突变相关。喹啉类药物抗性影响因子尚不完全清楚。恶性疟原虫 5号染色体上的多药耐药基因 1及 7号染色体上的cg2基因可能是抗性相关基因 ,但二者都不能完全解释抗性 ,尚待深入研究。

【Abstract】 The acquired drug resistance remains a major challenge to successful chemotherapy of malaria. The molecular mechanism of resistance to the antifolates has been well characterisied and is due to structural changes in the target enzymes resulting from the point mutations of DHFR or DHPS gene. With respect to quinoline antimalarials, the factors of resistance have yet to be fully elucidated. Pfmdr1 gene on chromosome 5 and cg2 gene on chromosome 7 in Plasmodium falciparum are considered to be associated with the quinoline drug resistance, but the exact mechanism remains unclear. Extensive study is required to completely understand the quinoline drug resistance. [
