龟龄集对大鼠海马结构内突触小泡蛋白年龄变化的影响
EFFECTS OF GUI LINGJI ON AGE RELATED CHANGES OF SYNAPTOPHYSIN IN RATS’ HIPPOCAMPAL FORMATION
【摘要】 目的 :研究大鼠海马结构内突触小泡蛋白年龄变化的影响。方法 :实验用 Wistar大鼠、免疫组织化学结合图像分析法。结果 :2 4月龄大鼠齿状回多形层、海马 CA3辐射层内突触小泡蛋白免疫反应产物的灰度值明显大于 18月龄鼠 ( P<0 .0 0 1,P<0 .0 0 5) ;12月龄鼠经 6和 12个月服用龟龄集后 ,齿状回多形层、海马 CA3辐射层内突触小泡蛋白免疫反应产物的灰度值均比同龄对照组显著降低 ( P<0 .0 5)。结论 :大鼠海马结构内突触小泡蛋白随着年龄的增加而减少 ;服用龟龄集可延缓神经元的衰老 ,防止海马结构内突触小泡蛋白的丢失 ,改善学习记忆功能
【Abstract】 Objective: To study age related changes of synaptophysin in rat’ hippocampal formation and Guilingji’s influence on it. Methods: Immunohistochemistry combined with image analysis was used. Results: The gray density of immunoreactive products of synaptophysin in rats’ polymporph layer dentate gyrus and hippocampus stratum radiatum of CA 3 area of 24 month rat’s was obviously higher than that of 18 month one(P<0.001, P<0.005). 12 month rats, after taking Guilingji for 6 and 12 months respectively, their synaptophysin immunoreactive products gray density decline remarkably than the control groups of the same age(P<0.005). COnclusion: The synaptophysin in rats’ hippocampal formation decrease with growing; taking Guilingji can delay neuron aging and prevent synaptophsin in hippocampal formation from decreasing.
