β-淀粉样蛋白25-35对培养的大鼠隔区神经元存活和生长的影响
EFFECTS OF β-AMYLOID PEPTIDE(25-35) ON SURVIVAL AND GROWTH OF THE NEURONS OF SEPTAL AREA IN CULTURE
【摘要】 目的:探讨β-淀粉样蛋白对隔区神经元的作用。方法:采用神经细胞培养技术、MTT法、免疫细胞化学及图像分析技术,观察“老化”及正常的β-淀粉样蛋白25-35片段对隔区神经元存活、生长及胆碱能神经元胞体中胆碱乙酰转移酶活性的影响。结果:“老化”的β-淀粉样蛋白 20 μmol/L组的隔区神经元光密度值及突起长度比对照组,10μmol/L组及未“老化”的20 μmol/L组均明显减小;胆碱能神经元的突起长度及胞体平均灰度在“老化”μ-淀粉样蛋白 20 μmol/L作用下也均比其它组明显降低。结论:“老化”的μ-淀粉样蛋白 25-35对隔区神经元的存活和生长发育及胆碱能神经元中胆碱乙酰转移酶的活性有抑制作用。
【Abstract】 Objective:To study the effects of β-amyloid peptide(25-35) on the neurons of septal area in culture. Methods: Survival and growth of the neurons were observed by using MTT assay and immunocytochemistry staining; the mean grey degree of ChAT-positive neurons was examined by image analysis system. Results: MTT assay showed that average OD values of ’aged’ Aβ25-35 (20 μmol/L) group was lower than that of control, ’aged’ Aβ25-35 (10 μmol/L) group and Aβ25-35(20 μmol/L) group(p<0. 05). The morphological changes showed that average neurite length in ’aged’ Aβ25-35 (20 μmol/L) group was decreased remarkably than that of other 3 groups(P<0. of) and the average neurite length and average grey degree of ChAT-positive neurons were also decreased significantly in ’aged’ Aβ25-35(20 μmol/L) group. Confusion: ’Aged’ Aβ25-35 has the neurotpxic effects on cultured neurons of septal area.
