肝树突状细胞与骨髓树突状细胞体外生物免疫特性的比较研究

【摘要】 目的 探讨肝树突状细胞(hepatic dendritic cell,HDC)的生物主疫功能及在肝移植耐受中的作用。方法 分别从肝脏和骨髓分离、培养DC,以免疫组织化学及流式细胞术检测两种DC生物特性及免疫活性的差异。结呆 培养6~8d,骨髓 DC(BM-DC)有大量细胞从集落中释放,并呈成熟状态,细胞表面组织相容性抗原 MHC class、MHC class Ⅱ及共刺激分子CD80(B7-1)、CD86(B7-2)都有较高水平表述,混合淋巴细胞反应(MLR)中表现出有强烈刺激T细胞增生的能力;而同时期的HDC具有非成熟DC的形态及功能特征,表面分子表达很低,无刺激T细胞增生的能力。结论 实验结果提示HDC具有BM-DC不同的生物特性可能是产生肝脏移植受特性的一个重要原因。

【Abstract】 Objective To investigate the biological characteristics of hepatic dendritic cell(HDC) and its functions in the immune tolerance of hepatic allografts. Methods DC progenitors were respectively isolated from liver and bone marrow of C57BL/6 J mice, and were propagated in liquid culture with GM-CSF. The immune function was detected by using immunochemical staining, FCM and MLR. Results After 6-8 days culture, most BM-DC released from proliferating aggregates. They expressed high level MHC class Ⅱ and coutimulating molecules CD80 (B7-1 ), CD86(B7-2 ), and the ability for strongly stimulating T cell proliferation, which showed the strong mature features. But HDC expressed lower level of those molecules, and failed in inducing primary allogeneic responses of naive T cells, Conclusion These observations demonstrated that liver-derived DCs have the bio-function of immature DC, which may con- tribute to the immune tolerance of hepatic allografts,
