大鼠脑内一氧化氮痛觉调制机制的实验观察

【摘要】 目的 探讨脑内一氧化氮在大鼠痛觉调制中的作用。方法 以钾离子透入引起大鼠甩尾反应的电流强度作为痛反应指标 ,观察侧脑室注射NG-硝基 -L -精氨酸甲酯、NG 硝基 D 精氨酸甲酯、硝普钠和血红蛋白等 ,对大鼠痛阈的影响。结果 侧脑室注射一氧化氮合酶抑制剂NG-硝基 -L -精氨酸甲酯后 ,大鼠痛阈明显增加 ;而注射NG-硝基 -D -精氨酸甲酯则对痛阈无明显影响。侧脑室注射一氧化氮供体物质硝普钠后 ,痛阈明显降低 ;而注射硝普钠与血红蛋白的混合液和Hb则对痛阈无显著影响。结论 大鼠脑内NO水平降低有镇痛作用 ,而NO水平的升高则有痛敏作用。提示 ,大鼠脑内NO在痛觉调制的复杂过程中 ,发挥着重要作用。

【Abstract】 Objective To Study of the effect of nitric oxide(NO) on pain modulation in the brain of rats.Methods The potassium inotophoresis induced tail-flick was used to measure the pain threshold(PT) of rats.The changes of pain threshold were investigated by microinjecting N G-nitro-L-arginine methyl ester(L-NAME),N G-nitro-D-arginine methyl ester(D-NAME),sodium nitroprusside(SNP) and hemoglobin(Hb) into the cerbral ventricle of rats.Results The pain threshold of rats was increased significantly by microinjecting L-NAME into the ventricle of brain.Significant hyperalgesic effects were observed by injecting SNP into the cerebral ventrile of rats,while the pain threshold showed no change by injecting the mixed liquid which included SNP and Hb.Conclusion The NO in the brain may be involved in pain modulation.An increase of NO in the brain produces hyperalgesia,and a decrease of NO level produces analgesia.
