双水相非离子表面活性剂萃取法分离胆固醇氧化酶
Separation of Cholesterol Oxidase by a Nonionic Surfactant- based Aqueous Two-phase Extraction System
【摘要】 TritonX -114是一种常用的表面活性剂。双水相萃取分离法适合于分离具有一定疏水特性的水溶性蛋白质。在研究了其浊点、浓度、pH值、分离时间和分离温度等影响因素后 ,得到了分离胆固醇氧化酶的优化条件。可以直接对发酵上清液进行处理 ,一次萃取可使酶液体积缩小10倍 ,总酶活回收率为70%。
【Abstract】 Nonionic Surfactant- based Aqueous Two-phase System is suitable for extraction of soluble protein which has the hydrophobic groups on its surface. Using this method for separation of cholesterol oxidase, the optimized separation condition were determined on the basis of the studies of the Triton X-114 cloud point, surfactant concentration, pH, mixture time and temperature. In only one step of clarification, more than 70% of the enzyme could be extracted and the volume was decrease into 10%.
