【摘要】 目的 :建立免疫印迹法检测血清抗Sa抗体 ,并对其敏感性、特异性和稳定性进行评价。方法 :通过 3组正交设计实验 ,确立检测方法的最佳条件 ;主要条件为硝酸纤维素膜应用 1 5%小牛血清封闭 2h ,被测血清作 1 :40稀释 ,酶标抗体作1 :80 0稀释。结果 :抗Sa抗体对类风湿性关节炎 (RA)敏感性为 42 .7% ,特异性为1 0 0 % ;结论 :建立的免疫印迹法检测血清抗Sa抗体结果较稳定 ,并有较高特异性更多还原

【Abstract】 Objective:To establish a method for detecting anti Sa antibody in serum by western blot and evaluate its sensitivity,specificity and stability.Methods:We established the best conditions for detecting anti Sa antibody by 3 orthogonal design tests.Results:The main conditions are as follows:The nitrocellulose membrane was blocked in 15 % bull serum solution for 2 hours;the patients′serum was diluted at 1:40 while labeled antibody at 1:800;the sensitivity of anti Sa antibody to rheumatoid arthritis was 42.7 % and specificity was 100 %.Conclusion:The results detected by western blot are stable and specific.更多还原
