卡氏肺孢子虫肺炎动物模型的建立及其检测

【摘要】 目的 为开展人卡氏肺孢子虫的病原学形态观察、免疫化学、分子生物学特性以及药物治疗的研究建立卡氏肺孢子虫肺炎大鼠模型。方法 给 SD大鼠腹股沟皮下注射醋酸可的松 ,6~12周后 ,收集支气管肺泡灌洗液和制作肺组织印片 ,组化染色 ,镜下观察。结果 实验大鼠的肺印片肺孢子虫病原学染色阳性率为 96 % (2 4/ 2 5 ) ,在 2 4只肺孢子虫病原学染色检查阳性的大鼠中 ,11只大鼠 BAL s沉淀涂片阳性。本次诱导实验包囊检出时间最早的为用药 40天 ,且随用药时间的延长 ,感染程度逐渐加重。对照组均未检出阳性。结论 建立了卡氏肺孢子虫肺炎大鼠模型。

【Abstract】 Objective To establish pneumocystis carinii  penumonia(PCP) rat model and detect human PCP. Methods SD rats were immunosuppressed with biweekly injection of cortisone acetate (25 mg) for 6~12 weeks and then killed. The development of P. carinii lung infection was monitored through giemsa, Weigert Giemsa and Chalvardjian’s toluidine blue O stainings of lung imprints and brochial aloveolar lavage fluids (BALFs). Results P. Carinii was observed in 24 of 25 experimental SD rats in lung imprints, among which 11 were positive in BALFs. Conclusions SD rat model of PCP has been successfully developed in our experiment. And the induction rate is similar to those reported abroad and higher than results of some domestic reports and has achieved our prospective.
