一氧化氮与实验性急性胰腺炎关系的探讨

【摘要】 目的 :探讨一氧化氮 (NO)在急性胰腺炎 (ANP)中的作用。方法 :采用胰管结扎及加压注射法建立大白兔急性胰腺炎模型 ,并采用还原型尼克酰胺嘌呤二核苷酸脱氢酶 (NADPH -d)与四唑仑硝基兰 (NBT)组成的孵育液对胰腺组织进行一氧化氮合酶 (NOS)的组化染色 ,以观察NO的变化。结果 :经Winslow氏法测定动物血清淀粉酶为 ( 1 3 1 8± 1 98.2 )u/L(实验组 ) ,而对照组为 ( 56± 1 8.6)u/L。并经生化分析及组织学检查提示急性胰腺炎 (SAP)动物模型建立。NOS组化染色显示 ,SAP模型组胰腺的腺泡细胞被染成深蓝色 ,而对照组被染成浅蓝色 ,染色强度SAP模型组明显高于对照组 ,结果表明NO与SAP具有相关性。结论 :NO参与SAP的病变 ,并起重要作用

【Abstract】 Objective:To inquire into the effects of nitric oxide(NO) on the pathological process of serious acute pan- creatitis(SAP).Methods:The SAP model of rabbit is constructed by ligation of pancreatic duct and inter-duct pressure injection.After construction of the SAP model,the nitric oxide synthase(NOS) of the pancreas is histochemically stained with incubation solution composed of NADPH-d and NBT,and the changes of NO levels are investigated.Results:The amylase concentrations (determined by Winslow method) of the experimental group and control group are (1318±198.2)u/L and (56±18.6)u/L respectively.Biochemical analysis and histological examination demonstrate that the SAP model is successfully constructed.The histochemical stain of NOS demonstrates that acinar cells of pancreas in the experimental group are stained to dark blue and those in the control group are light blue.The stain of the experimental group is obviously more intensive than that of the control group.Conclusion:NO participates in the pathological process of SAP.
