外源基因在转基因油菜后代中的表达及遗传学分析

【摘要】 以转基因油菜后代 ( R1 和 R2 代 )株系为材料 ,系统地研究了苏云金杆菌杀虫蛋白基因Cry A和选择标记基因 npt 导入油菜后的遗传规律、表达强度和抗虫效应 .结果表明 :R1 代卡那霉素抗性为 3∶ 1分离方式 ,在 R2 代卡那霉素抗性植株中 ,有三分之一为 npt 基因纯合型( T∶ T) ,另三分之二为杂合型 ( T∶O) ,其遗传方式符合孟德尔单因子显性遗传 ;转化植株可耐受1 50 mg/ L那卡霉素的选择压力 ;约一半的转化植株具有杀虫或抑虫效力 ,幼虫校正死亡率最高可达 78% .

【Abstract】 We studied expression and inheritance of foreign Cry IA npt Ⅱ genes in transgenic progenies of oilseed rape ( Brassica napus and B.juncea ) in the present paper. The results showed that transgenes were delivered in the transgenic progenies according to Mendelian Law of single gene segregation (3∶1).Kanamycin resistance and insect resistance were maintained in R 1 and R 2 generations of the transgenic oilseed rape progenies. There were 1/3 homozygotes and 2/3 heterozygotes in the R 2 kanamycin resistant plants, and some of the plants could grow under high selective pressure (over 150 mg/L kanamycin).Half of the transgenic R 1 and R 2 plants exhibited tolerance to pest insects such as Laphygma exigua, and mortality of the larvae was about 44%~78% in the leaf feeding experiments.
