红豆杉细胞的筛选培养基优化
Optimization on the Sifting Medium from Taxus Chinensis
【摘要】 运用正交试验优化小细胞团法筛选红豆杉细胞系的筛选培养基 .培养基为改良的MS培养基 ,培养基质量浓度为硝酸钾 1 90 0mg·L-1,硝酸铵 82 5mg·L-1,磷酸氢二钾 1 70mg·L-1,蔗糖质量浓度 30 g·L-1,葡萄糖质量浓度 5 g·L-1,并且添加适量的酵母提取物、甘氨酸、维生素C、叶酸、氯化胆碱、泛酸钙等 .该培养基明显提高了细胞生长率 ,并能增加紫杉醇的含量
【Abstract】 Taxus chinensis cell line is selected by a small cell aggregate culture, and its plate culture medium is studied. The minimum size of inoculum for callus growth is 14.5?g (cell dry weigh)/L medium, and the growth cycle of callus is about 35 days. Improved MS Medium is suitable for callus growth. The cell growth rate can be promoted by increasing the glucose concentration to 5?g·L -1 , sucrose to 30?g·L -1 , and also by supplementing with yeast extract, glycine, Vitamin C, folic acid, choline chlorice and calcium pantothenate. The taxol content can also be promoted by the above treatments.
