生物体内泛醌的种类及合成条件的探讨
Study on ubiquinone species and synthetic condition for organisms.
【摘要】 通过薄层层析、高效液相色谱等方法对大肠杆菌、酿酒酵母、粟酒裂殖酵母和烟草等四种不同生物所含的泛醌 (即辅酶 Q)进行了定性分析 ,确定了各种生物所含的泛醌种类 .并利用表达泛醌合成基因的大肠杆菌培养过程中泛醌含量的变化以及添加前体物质对提高大肠杆菌泛醌合成量进行了初步研究 ,确定了稳定生长前中期是生物体中泛醌含量最高时期 ,引进泛醌合成的外源基因以及在培养基中添加 0 .1m M前体物质 -对羟苯甲酸可以提高生物体内泛醌的含量
【Abstract】 Ubiquinone, extracted from \$Escherichia coli, Saccharomyces cerevisiae, Schizosaccharomyces pombe\$ and \$Nicotiana tabacum\$, were analyzed by thin\|layer chromatography (TLC) and high performance liquid chromatography (HPLC), and the ubiquinone species of them were confirmed. Then, ubiquinone biosynthetic genes were expressed in \$E. coli\$. Effect of cultivative time and precursor \$p\$\|hydroxybenzoate on ubiquinone yield was investigated. The results show that the higher level of ubiquinone is produced in the stationary phage of growth period, and over\|expressing exogenous genes as well as adding 0.1mM \$p\$\|hydroxybenzoate may increase ubiquinone yield of organism.
