抗CD4人-鼠嵌合抗体的生物学特性及作用机理探讨
The biological characterization and mechamism of antiCD4 human/murine antibodies.
【摘要】 目的 比较抗 CD4人 -鼠嵌合抗体与鼠源性单抗在单向混合淋巴细胞反应 (ML R)和兔皮内移植物抗宿主反应(GVHR)中的作用 ,及嵌合抗体的抗体依赖性细胞介导的细胞毒 (ADCC)和补体依赖细胞毒 (CDC)活性 ,以进一步探讨抗CD4嵌合抗体的增殖抑制作用机理。方法 用四甲基偶氮唑盐比色分析法检测抗 CD4抗体在单向 ML R中的抑制作用。以免疫抑制剂处理后的大白兔作为 GVHR的动物模型 ,对抗体的增殖抑制作用进行体内实验。结果 抗 CD4抗体对同种细胞性抗原诱导的外周血单个核细胞增殖反应有明显的抑制作用 ,能明显抑制 GVHR的阳性反应 ,嵌合抗体的抑制作用较鼠源性单抗强。抗 CD4嵌合抗体具有 ADCC效应 ,而无 CDC效应。结论 抗 CD4抗体可能通过多种抑制机制下调 CD4+ T细胞介导的免疫反应 ,具有有效的免疫抑制作用
【Abstract】 Objective To compare the effects of anti CD4 humua/murine antibodies and murine McAbs on one way mixed lymphocyte reaction(MLR)and rabbit intracutaneous graft versus host reactin(GVHR),and to measure the ADCC and CDC activity of chimeric antibodies.Methods Using MTT method,the inhibitory effects of anti CD4 antibodies on one way MLR was studied,and by using immunosuppressant treated rabbit as GVHR animal model,the in vivo experiment of antibodies inhibitory effect on proliferation was performed.Results Both anti CD4 human/murine chimeric antibodies and murine McAbs could obviously inhibit the PBMC proliferation induced by allogeneic cells antigen and positive reaction of GVHR.The inhibitory effect of anti CD4 chimeric antibodies was stronger than that of murine McAbs.Chimeric antibodies could mediate ADCC against CEM cell line,but poorly lytic in complement dependent cytotoxicity assays(CDC).Conclusion Anti CD4 antibodies have an active immunosuppressive effect by various inhibitory mechanisms to down regulate CD4 + T mediated immunoreaction.
