17α-羟孕酮对离体培养的对虾卵巢组织发育的促进作用
EFFECT OF 17α-HYDROXYPROGESTERONE ON IN VITRO CULTURE OF OOCYTES OF SHRIMP (Penaeus Chinensis)
【摘要】 报道了用3种不同浓度的17α─羟孕酮体外培养中国对虾卵巢组织的试验结果。结果显示,在对虾卵巢组织培养基中添加2×10-6~2×10-8μg/ml的17α─羟孕酮溶液,对对虾卵巢细胞分生和卵母细胞直径增大具有显著或极显著的刺激作用,在相同培养时间内,试验组卵母细胞直径比对照组增大30%~50%。随着激素浓度的增加卵原细胞分生和卵母细胞直径增长加快。体外培养的对虾卵母细胞最大直径可达160μm,外层没有形成滤泡细胞包被,继续培养,卵膜破裂,内涵物外露,细胞裂解,无法形成成熟的卵母细胞。
【Abstract】 The experimental results in vitro stimulation of oocytes of shrimp (Penaeus chinensis) by 17α-hydroxyprogesterone at concentration of 2×10-6, 2×10-7 and 2×10-8 μg/ml are reported. The results showed that the hormone had significant effect on the oocytes development. Under identical culture condition, the mean oocyte diameters in the experimental groups with hormone were 30%~50% larger than that in contrast group. With increase of hormone concentration, reproduction of oocyte quickened and oocyte diameter increased. Through a week culture,the mean diameters of oocytes in the experimental groups were 85.1 ±12. 4, 102.7±11. 5 and 131. 4±12. 2μm respectively, while that in contrast group was 67. 7±9. 2 μm.The biggest oocyte was about 160μm in diameter, and there were no follic1e cells around the surface of it. The membrane of the oocyte would burst and the contents exposed, if it was cultured continuously.
