巴巴多斯芦荟多糖对内毒素化大鼠肾功能衰竭的保护作用

【摘要】 目的 :研究巴巴多斯芦荟多糖 (AP)对内毒素化大鼠肾功能衰竭的保护作用。方法 :分别采用一次性静脉注射 2 50 μg·kg- 1 和连续 7天每天一次腹腔注射 50 μg·kg- 1 K0 3肺炎杆菌内毒素 (LPS) ,建立大鼠急性及慢性肾功能衰竭模型。以通过测定血清肌酐、尿素氮和菊糖肾清除率评价静脉及连续腹腔注射AP对内毒素所致大鼠肾衰的保护作用。结果 :静脉注射AP 2 5mg·kg- 1 可使内毒素引起的大鼠菊糖肾清除率的下降由 30 1 3 %恢复到2 1 36 % ;腹腔注射AP 2 5mg·kg- 1 、1 0mg·kg- 1 均可显著降低内毒素引起的大鼠血尿素氮和血肌酐水平升高 (P <0 0 1、P <0 .0 5)。结论 :AP对K0 3肺炎杆菌内毒素所致大鼠肾功能衰竭具有一定的保护作用

【Abstract】 OBJECTIVE:To investigate the protective effects of Barbandensis aloe miller polysaccharide (AP) on the renal failure by Klebsiella pneumoniae 03 lipopolysaccharide (LPS) in rats. METHODS: The rats were treated respectively with single dose of 250 μg·kg 1 LPS intravenously and 50 μg.kg 1 LPS interpenetratively per day for 7 days to establish the acute and chronic renal failure models.After an intravenous administration of 100 mg.kg 1 inulin,the renal clearance of inulin was estimated for each rat with one compartmental method. Inulin was measured by the standard colorimetric method. The concentrations of serum urea nitrogen and creatinine were measured by using commercially available kits.RESULTS:The results show that LPS treated rats decreased the renal clearance of inulin significantly. The decreased rate of renal clearance recovered from 30.13% to 21.36% after the treatment of AP.In chronic renal failure experiment, AP showed to inhibit the increase of serum Cr and BUN in LPS treated rats. It could be concluded that AP has obvious protective role to renal function in endotoxinic rats.
