部分背根切断对备用背根节trkC表达的影响

【摘要】 为探讨去部份背根(切除一侧L1-L5, L7-S2背根节,保留L6背根为备用根)猫备用L6背根节(DRG)trkC表达的变化,用特异的trkC兔抗血清以免疫组织化学方法染片,观察trkC在DRG的分布。计数正常组一侧,术后3天及10天组手术侧L6 DRG大、小神经元trkC的阳性细胞数,结果用q检验进行统计分析。结果:trkC的免疫阳性反应(trkC-IR)主要分布于DRG的大细胞及少数小神经元胞浆内。部分去背根后3天, L6 DRG大、小 trkC-IR阳性神经元数与正常组者比较无显著变化(P>0.05)。但10天组者,trkC阳性大神经元的数量明显减少,而小神经元数则明显增多(P<0.05),表明部分背根切断使备用DRG trkC的表达在大神经元减少,而小神经元则增加。以上提示,备用DRG trkC的表达变化可能与脊髓可塑性有关系。

【Abstract】 This study sought to explore the expression of trkC in spared dorsal root ganglion(DRG) following unilateral dorsal root rhizotomy. 15 cats were div ided into three groups: the intact group, the 3-day and 10-day groups after op eration (unilaterral L1-L5 DRG and L7-S2 DRG were sectioned, but L6 DR G was spared). The animals were sacrificed on the 3rd day and 10th day after oper ation, respectively. The L6 DRG from the three groups were taken and cut into frozen sections (20μm). The sections were stained using specific antiserum for trkC by th e immunohistochemical ABC method. The numbers of large sized neurons (>57 μm ) and small sized neurons (<42μm) for trkCIR were counted respectively. The re sults showed that the number of positive large sized neurons for trkC of the 10d ay group after operation decreased significantly than that of the intact group a nd the 3-day group after operation, while the number of positive small sized ne urons for trkC of the 10-day group increased apparently than that of the intact group and the 3day group after operation (P<0.05). This experiment demons trated that unilateral partial dorsal root rhizotomy could change the expression of t rkC might in large neurons and small sized neurons, and the results suggested th at trkC might involve in the spinal cord plasticity following partial dorsal root rhi zotomy.
