2株分离的嗜人T细胞白血病病毒I型的PCR检测
Human T-Cell Leukemia Virus Type I Detected by Polymerase Chain Reaction
【摘要】 [目的 ]用 PCR法检测嗜人 T细胞白血病病毒 I型 (HTL V- I)的分离情况。 [方法 ]采集 2例日本鹿儿岛县成人 T淋巴细胞白血病 (AL T)病人的肝素抗凝血 ,分离外周血单核细胞 (PBMCs) ,加入植物血凝素和白介素 - 2 ,与健康人 PBMCs共培养进行 HTL V- I的分离 ,然后抽提 DNA,用 HTL V- I的 gag1和 tax1两引物进行 PCR扩增 ,对扩增产物进行琼脂糖凝胶电泳确定病原分离结果。 [结果 ]2株都扩增出了 HTL V- I前病毒 DNA的 gag1和 tax1片断 ,证明了嗜人 T细胞白血病病毒 I型的分离成功。 [结论 ]PCR法是快速、准确检测嗜人 T细胞白血病病毒感染的有效方法之一。
【Abstract】 Objective] To study on isolating Human T cell Leukemia virus type I (HTLV I) by polymerase chain reaction (PCR) [Methods] PBMCs from 2 heperinized blood specimens of ATL patients were collected and co cultured with peripheral blood monocytes (PBMCs) donated by uninfected healthy donors with the presence of phytohemagglutinin (PHA) and interleukin 2 (IL 2) HTLV I proviral DNA was prepared from CD4 + T cell by using the DNA extractor kit The proviral DNA was amplified by PCR using primers gag1 or tax1 The amplified products were detected with agarose gel electrophoresis [Results] We detected the 236bp bands of gag1 and 1058bp bands of tax1 by agarose gel electrophoresis and proved to separate the HTLV I provirus successfully [Conclusion] PCR is one of the useful methods to detect the infection of HTLV I
