不同剂型的蝎毒抗癌多肽对小鼠肝癌的抑制作用及带瘤小鼠免疫功能的影响

【摘要】 目的 :观察蝎毒抗癌多肽 (antineopasticpolypeptidefromButhusmartensiivenom ,APBMV)对小鼠肝癌 (hep atoma 2 2 ,H2 2 )的生长抑制作用及带瘤小鼠免疫功能的影响。方法 :①将接种H2 2 细胞 2 4h后的带瘤小鼠 45只随机分为非正常对照组、5 Fu组、APBMV针剂组和胶囊组 4组 ,给予不同的药物处理 1 0d后 ,观察瘤块及脾脏重量的变化 ,计算抑瘤率和脾重指数 ;②接种H2 2 细胞 2 4h后的带瘤小鼠 40只随机分为非正常对照组、5 Fu组、APBMV针剂组和APBMV针剂 +5 Fu组 ,给予APBMV针剂和 /或 5 Fu 5d后 ,分别观察带瘤小鼠NK细胞活性及淋巴细胞转化率。结果 :① 5 Fu和APBMV针剂组对H2 2 的抑瘤率分别达 47.81 %和 40 .0 7% ,与非正常对照组相比差异均有显著性 (P <0 .0 5 )。APBMV针剂及胶囊组的脾重指数分别为 70 .0 2mg/1 0g和 71 .2 2mg/1 0g,与非正常对照组相比P <0 .0 1 ;②APBMV针剂组及APBMV针剂 +5 Fu组带瘤小鼠的NK细胞活性明显增强 ,淋巴细胞转化指数明显增高(P <0 .0 5 )。结论 :APBMV针剂可抑制H2 2 的生长 ,并增强带瘤小鼠机体的免疫功能

【Abstract】 Aim: To observe the influence of antineoplatic polypeptide from Buthus martensii venom (APBMV) on the growth of hepatoma 22(H 22 )and the immunity of H 22 bearing mice. Methods: ①The 45 mice inoculated by H 22 cells for 24 hours were divided into 4 groups:non normal control group,5 Fu treated group,APBMV injection and capsule groups. H 22 bearing mice were treated by different chemicals for 10 days and the weight changes of H 22 tumor and spleen were observed. The tumor inhibitory rate and spleen weight index were calculated. ②The 40 mice inoculated by H 22 cells for 24 hours were divided into 4 groups:non normal control group,5 Fu treated group,APBMV injection or/and 5 Fu groups. H 22 bearing mice were treated with APBMV injection or/and 5 Fu for 5 days. The activity of NK cell and the transformation rate of lymphocytes were observed. Results: ①Inhibitory rates in 5 Fu and APBMV injection groups were 47.81% and 40.07%, respectively. Compared with the non normal control group, the differences were significant ( P <1.05). The spleen weight indexes were 70.02 mg/10 g and 71.22 mg/10 g in APBMV injection and capsule groups, respectively. Compared with the non normal control group, the differences were significant ( P <0.01). ②The activity of NK cell was enhanced remarkably and the transforming rate of lymphocytes was increased evidently in both APBMV injection group and APBMV injection+5 Fu group ( P <0.05). Conclusion: APBMV could inhibit the growth of H 22 and strengthen the immunity function of H 22 brearing mice.
