蝎毒抗癌多肽纯化组分Ⅲ对小鼠肝癌的生长抑制作用及带瘤小鼠胸腺重量的影响

【摘要】 目的 :观察蝎毒抗癌多肽纯化组分Ⅲ (antineoplasticpolypeptide ⅢfromAPBMV ,AP Ⅲ )对小鼠肝癌 (hep atoma 2 2 ,H2 2 )的生长抑制作用及对带瘤小鼠胸腺重量的影响。方法 :将接种H2 2 细胞 2 4h后的昆明种小鼠 6 5只 ,随机分为 5组 :5 Fu组 1 0只 (2 0mg/kg) ;APBMV组 1 0只 (0 .0 4mg/kg) ;AP Ⅲ实验组 (Ⅰ :0 .0 4mg/kg ,Ⅱ :0 .0 3mg/kg) ,每组 1 0只 ;非正常对照组 2 5只 (生理盐水 ) ,均腹腔注射给药 ,1次 /d ,连续给药 1 0d。于末次给药 2 4h后 ,颈椎脱臼处死小鼠 ,剥离肿瘤和胸腺 ,称重 ,并作数据处理。结果 :AP Ⅲ实验组和APBMV组对H2 2 有明显的抑制作用 ,与非正常对照组比较差异有显著性 (P <0 .0 1或P <0 .0 0 1 ) ;5 Fu组胸腺质量低于非正常对照组 ,而AP Ⅲ实验组和APBMV组则高于或接近非正常对照组。结论 :AP Ⅲ具有抑制H2 2 生长的作用 ,高剂量的AP Ⅲ可增加带瘤小鼠胸腺的重量

【Abstract】 Aim: To observe the inhibitory effect of antineoplastic polypeptide Ⅲ from APBMV (AP Ⅲ)on mouse hepatoma 22 (H 22 )and influence of AP Ⅲ on thymus gland weight of tumor bearing mice . Methods: Sixty five mice were inoculated by H 22 cells and 24 h later were divided into 5 groups:10 for 5 Fu group (20 mg/kg), 10 for APBMV group (0.04 mg/kg),10 for AP Ⅲ group Ⅰ(0.04 mg/kg), 10 for AP Ⅲ group Ⅱ(0.03 mg/kg) and 25 for non normal control group(saline). The mice were administered by intraperitoneal injection once a day for 10 days and then killed. The weight changes of tumor and thymus gland were observed accurately and statistically. Results: Growth of H 22 was inhibited markedly by AP Ⅲ at two dosages used in this experiment and APBMV, compared with non normal control group, the differences were significant ( P <0.01 or P< 0.001), and indexes of thymus gland were lower in 5 Fu group and higher or similar in two AP Ⅲ and APBMV groups. Conclusion: Growth of H 22 was efficiently inhibited by AP Ⅲ .The thymus gland weight of H 22 bearing mice was increased by high dosage of AP Ⅲ.
