【摘要】 目的 :探讨人前列腺癌细胞转移潜能与KAI1基因表达的关系。方法 :运用Northern印迹杂交检测KAI1基因在不同转移潜能人前列腺癌细胞中的表达 ,以甲基化分析和聚合酶链反应 单链构象多态性分析 (PCR SSCP)研究KAI1低表达的机制。结果 :在转移性人前列腺癌细胞系PC3和PC3M中不论其转移潜能强弱 ,KAI1mRNA表达均降低。甲基化分析未发现KAI1基因 5’ 启动子部位CCm5CGGG甲基化。PCR SSCP分析显示 :KAI1基因第 7外显子在PC3和PC3M均显示异常带 ,PC3M较弱。结论 :KAI1基因表达降低与人前列腺癌的转移有关 ,但其转移潜能的强弱可能不取决于KAI1的调控。KAI1基因的低表达与 5’ 启动子部位CCm5CGGG甲基化无关 ,可能与该基因的突变失活有关。

【Abstract】 Aim: To investigate the relation between metastatic potential of prostate cancer cells and expressions of KAI1 gene Methods: Expressions of KAI1 mRNA in human prostate cancer cell lines with different metastasis potentials were detected by Northern blot hybridization Mechanism of its expressions was studied by assessing methylation status and PCR SSCP Results: Expressions of KAI1 mRNA reduced in human prostate cancer cell lines PC3 and PC3M No presence of CC m5 CGGG methylation was observed in a position of the 5’ promoter region of KAI1 gene PCR SSCP analysis indicated the existence of band shift in exon 7 of KAI1 gene in PC3 and PC3M Conclusion: Reduced KAI1 expressions may be associated with enhanced metastatic potential in human prostate cancer and difference of metastasis potential does not depend on the control of KAI1 gene Reduced expressions of KAI1 mRNA may associate with mutation in exon 7 of KAI1 gene other than presence of CC m5 CGGG methylation in 5’ promoter region of gene
