紫杉醇对小鼠巨噬细胞免疫活性的影响

【摘要】 目的 :研究紫杉醇对小鼠巨噬细胞免疫活性的调节作用。方法 :按常规方法收集纯化 8~ 12周龄BALB/c雌性小鼠腹腔巨噬细胞 ,用RPMI16 40培养基培养 ,按所用药物紫杉醇 (Paclitaxel)不同浓度和 (或 )干扰素 γ(INF γ)进行分组并设对照组。一氧化氮 (NO)测定采用Griess法。结果 :一定浓度的紫杉醇能够诱导巨噬细胞产生NO ,且随浓度增大而增加 ;紫杉醇联合INF γ时 ,诱导作用明显增加 ,且紫杉醇腹腔给药更易达到有效活化巨噬细胞浓度。结论 :腹腔给药有利于紫杉醇发挥抗肿瘤作用 ,若与INF γ联合更能发挥其免疫化学治疗作用

【Abstract】 Aim:To study the regulation of Paclitaxel to macrophage activation.Methods:8~12 weeks old BALB/c female mice were used.Peritonal macrophages were collected and purified as reported previously and cultured in RPMI 1640 culture.Peritonal macrophages were divided into groups according to the reagents(Paclitaxel and/or INF γ).Peritonal macrophages without reagents were served as control group.Nitric oxide concentration in macrophages derived supernatants was measured by Griess method.Results:Paclitaxel with certain concentration could induce macrophages to produce nitric oxide and the effect became strong with concentration increasing.Paclitaxel showed stronger effect on macrophages in combined with INF γ. Conclusion:Using Paclitaxel in peritonal cavity could get effective concentration of activating macrophages easily.
