棉花DNA导入苎麻的RAPD验证
RAPD Assay of Cotton DNA Introduced into Ramie
【摘要】 为了寻找通过超干胚浸泡法已经将湘棉 12号 DNA导入苎麻湘苎 3号基因组中的分子证据 ,对所产生的变异后代进行了 RAPD验证 .所用 80个随机引物中 ,有 6个引物检测出变异材料 97- 2 4与受体的 DNA多态性 ,3个引物检测出变异材料 97- 2 8与受体的多态性 ,4个引物检测出变异材料 97- 4与受体的多态性 ,并且有 2个引物在变异后代中扩增出供体特异带 .这些结果说明棉花 DNA导入苎麻后引起了后代基因组的变异 .
【Abstract】 Offsprings derived from introducing cotton “Xiangmian 12” DNA into ramie “Xiangzhu 3” through soaking superdried seed embryo were assayed with RAPD. Of the 80 arbitrary primers used,products of 6 primers were detected to have polymorphism between offspring 97 24 and its recipient,and those of 3 between 97 28 and recipient,those of 4 between 97 4 and recipient,respectively. Meanwhile,there were 2 primers that amplified specific DNA bands only existing in offsprings and the donor. These results showed that cotton DNA induced the variances in genome of ramie.
