应用单细胞凝胶电泳研究镉对大鼠肝细胞DNA损伤的影响
Effects of Cadmium on DNA Damage in Rat Liver Cells,in Vivo and in Vitro Studies With Single Cell Gel electrophoresis
【摘要】 [目的 ]研究一定剂量的氯化镉对离体和在体大鼠肝细胞 DNA损伤的作用。 [方法 ]应用单细胞凝胶电泳或慧星试验。 [结果 ]在 2 .185μmol/ L、 4 .3 75μmol/ L、 8.75μmol/ L、 17.5 0μm ol/ L、 3 5 .0 0μmol/ L的剂量条件下 ,氯化镉均可引起离体大鼠肝细胞 DNA损伤。 5μmol/ kg、 10μmol/ kg、 2 0μmol/ kg的氯化镉也可引起在体大鼠肝细胞 DNA损伤。氯化镉引起的离体和在体大鼠肝细胞 DNA损伤均存在明显的剂量 -反应关系。 [结论 ]一定剂量的氯化镉能引起离体和在体大鼠肝细胞 DNA损伤。
【Abstract】 To study the effects of cadmium chloride on DNA damage of rat liver cells in vivo and in vitro Single cell gel electrophoresis or Comet assay was used At the concentration of 2 185 μmol/L, 4 375 μmol/L, 8 75 μmol/L, 17 50 μmol/L, 35 00 μmol/L, cadmium chloride could induce DNA damage of rat liver cells in vitro, and at the doses of 5 μmol/kg, 10 μmol/kg, 20 μmol/kg, cadmium chloride could induce DNA damage of rat liver cells in vivo respectively The in vivo and in vitro results also showed the obvious dose response relationship between the rates of Comet cells and the doses of cadmium chloride [Conclusion]The research infered that at certain dose, cadmium could induce DNA damage and had toxic effects on rat liver cells
