用MCF-7细胞增殖法研究国产多氯联苯的类雌激素效应

【摘要】 用人乳腺癌细胞系MCF7细胞增殖试验,对两种国产多氯联苯混合物PCB3和PCB5的细胞增殖生物效应进行了研究.和典型雌激素17β雌二醇相比,PCB3在78pg/mL和182pg/mL水平表现出其类似的生物效应,其相对的细胞增殖效应分别为094和086.但在较高浓度91ng/mL,细胞增殖效果为阴性.PCB5和PCB3类似,在较低水平83pg/mL,其相对的细胞增殖效应为107,较高水平(166pg/mL以及83ng/mL时)细胞增殖效应为阴性.

【Abstract】 Recently,more evidences showed that some PCB mixtures might possess estrogenic effect. In this paper, the E\|Screen method was used to determinate the proliferation effects of two Chinese commercial PCB mixtures, China PCB\-3 and China PCB\-5.At the levels of 7\^8?pg/mL and 182?pg/mL, China PCB\-3 showed 0\^94 and 0\^86 of relative proliferation effect (RPE) compared to those of 17β\|estradiol(its RPE is 1\^00)respectively.At the level of 8\^3?pg/mL,China PCB\-5 acted nearly the same as China PCB\-3,its RPE was as high as 1\^07 compared to that of 17β\|estradiol.However, China PCB\-3 did not express cell proliferation effect at higher levels of 9\^1?ng/mL for China PCB\-3 and of 166?pg/mL and 8\^3?ng/mL for China PCB\-5 respectively.
