【摘要】 将链霉菌菌株Streptomycessp .纯化后 ,测得其葡萄糖异构酶的比活力为 0 .4 1 4u/mL ,使用苯酚氯仿法抽提到的基因组DNA浓度较低 ,RNA和蛋白质较多 ,但用大量法时仍可得到大量的DNA .使用试剂盒法得到的基因组DNA浓度较大 ,但仍有大量RNA存在 .由亚精胺法得到的基因组DNA浓度较好 ,RNA很少 .自行设计的改进法得到的基因组DNA ,量特别大 ,纯度很高 ,DNA大小集中在 3 0kb左右 ,RNA很少 ,非常适用于相应的基因工程操作 .更多还原

【Abstract】 Megathermal Streptomyces sp.was purified and its glucose isomerase activity is 0.414 u/mL.Low concentration genome DNA can be extracted by method of phenyphenol-chloroform,but there are RNA and amount of p rotein.Large amount of genome DNA can be gotten using large-scale method.High co ncentration genome DNA can be extracted by method of kit ,still with amount of R NA.High concentration of genome DNA with little RNA, can be extracted by method of spermidine.From self-designed and improved method,large amount of gen ome DNA with high purity and little RNA can be gotten,and the base number of mai n part of DNA is about 30 kb.It’s convenient to later gene engineering manipula tion.更多还原
